(S)-1,2,3,4-Tetrahydro-2-[3-methyl-N-[(phenylmethoxy)carbonyl]-L-valyl]-3-isoquinolinecarboxylicacid, methyl ester To a solution of Compound C (5.31 g, 20 mmol) in dichloromethane (80 mL) at 0C under argon were sequentially added diisopropylethylamine (10.6 mL, 60 mmol), benzotriazol-1-yloxytris- (dimethylamino)phosphonium hexafluorophosphate (5.08 g, 20 mmol) and (S)-1,2,3,4-Tetrahydroisoquinoline-3-carboxylic acid, methyl ester, hydrochloride (5.68 g, 25 mmol). The reaction mixture was allowed to warm to 5C over 2 hours, and then stirred overnight (16 hours). The mixture was washed with 1N hydrochloric acid, saturated sodium bicarbonate and brine (50 mL each). The organic layer was dried (magnesium sulfate), filtered and concentrated to afford an oil. Purification by flash silica gel column chromatography eluding with 20% ethyl acetate in hexanes afforded Compound D (4.0 g, 45%).
