A series of 1,2,3,4-tetrahydroisoquinoline derivatives were synthesized and evaluated for anticonvulsant activity against intracerebro-ventriculas (i.c.v.) N-methyl-D-aspartate (NMDA)-induced seizures in mice. Among these compounds, (+)-1-methyl-1-phenyl-1,2,3,4-tetrahydroisoquinoline hydrochloride ((+)-1a, FR115427) was the most effective anticonvulsant, and also protected CA1 hippocampal neurons from ischemia-induced neuronal degeneration in rats at 32 mg/kg i.p. In addition, (+)-1a showed anti-hypoxic activity in mice at 3.2-32 mg/kg i.p. The absolute configuration at the C-1 position of the isoquinoline ring was determined to be S by a single-crystal X-ray analysis of (+)-1a (+)-di-p-toluoyl-D-tartrate. Structure-activity relationships with regard to the anticonvulsant activity of this series of compounds are discussed, and the three-dimensional structures of (s)-(+)-1a and MK801 are compared.

This MOF can serve as coating material in gas chromatographic capillary column for the separation of some low boiling point essential oils. The prepared columns have good recognition ability and excellent selectivity toward a series of organic compounds, including alcohols, aldehydes, ketones, carboxylic acid, ethers, ester, and amines. It is found that the strained metal sites, van der Waals interactions, C-H?pi attraction, and weak nonclassical hydrogen bond contribute to the recognition and selectivity of prepared columns. The grand canonical Monte Carlo technique is used to simulate the interactions of the adsorbates with MOF. The calculated van der Waals energies agree with the results of gas chromatographic separation.

Discovery of novel N-sulfonamide-tetrahydroquinolines as potent retinoic acid receptor-related orphan receptor gammat inverse agonists for the treatment of autoimmune diseases

Targeting the nuclear receptor RORgammat is thought to be effective in autoimmune disorders. Tertiary sulfonamide 1 was found to be a potent RORgammat inverse agonist previously. However, the high hepatic clearance value limits its druggability. In this study, we designed and synthesized a series of N-sulfonamide-tetrahydroquinolines by molecular modeling and scaffold hopping strategy, aiming at improving the metabolic stabilities. Detailed SAR exploration led to identification of potent RORgammat inverse agonists such as 13 with moderate binding affinity and inhibitory activity of Th17 cell differentiation. Binding mode of 13 with RORgammat-LBD was revealed by molecular docking. Moreover, 13 showed lower intrinsic clearance in mouse liver microsomes compared with 1 and potent in vivo efficacy and safety in psoriasis models, which can be used as a good starting point for the further optimization.

Five new lanthanide tetrahydrofuran-2-carboxylate (THFC) complexes [Ln(THFC)2(H2O)2]¡¤L¡¤H 2O (Ln = Eu; L = Cl-, Br-, NO3 – and Ln = Tb; L = Cl-, NO3-) have been synthesized. The crystal structures of four compounds were solved by the X-ray diffraction method. The Ln3+ ions are coordinated to eight oxygen atoms: six oxygen atoms are provided by two tridentate bridging-cyclic THFC ligands, and the other two by water molecules. The Ln3+ coordination polyhedron can be described as a distorted square antiprism. The structures consist of planar layers linked by hydrogen bonds through outer-sphere L anions and H2O molecules. The influence of the L anion on the Eu3+ luminescence center is analyzed with the help of luminescence and IR vibrational spectroscopy. The distortions of the square antiprism increase with the size of the L outer-sphere anions. Consequently, the lowering of the symmetry of the crystal field around the Eu3+ ion appears as an extra splitting of the 7FJ levels under the influence of the crystal field components of the second and fourth ranks.

Conformational and steric aspects of the inhibition of phenylethanolamine N-methyltransferase by benzylamines

Compounds of the benzylamine (BA) class are potent inhibitors of phenylethanolamine N-methyltransferase (PNMT, EC 2.1.1.28). Restriction of the aminomethyl side chain through its incorporation into a cyclic framework as in 1,2,3,4-tetrahydroisoquinoline (THIQ) or 2,3,4,5-tetrahydro-1H-2-benzazepine (THBA) results in enhanced potency as an inhibitor, suggesting a conformational effect in the binding of BAs to the active site; however, these ring systems still retain a high degree of flexibility. We have synthesized a series of conformationally defined analogues of benzylamine in order to probe the effect of conformation, as well as the influence of steric bulk, on PNMT inhibition by this class of ligands. In addition, 1-, 3-, and 4-methyl-substituted THIQs were synthesized and evaluated as flexible models for steric bulk tolerance about this ring system. Substitution by a methyl group on either benzylic position of THIQ results in diminished activity as a PNMT inhibitor; however, 3-methyl-THIQ (11) shows enhanced activity as an inhibitor vs THIQ itself. Full conformational restriction of the BA side chain in analogues 4-8 results in a dramatic loss in inhibitor potency. We attribute this effect to a negative steric interaction between the alkyl bridging units above (or below) the heterocyclic ring systems and an active-site amino acid residue. Conformational restriction of THIQ employing a bridging unit that is not located above (or below) the ring system (9) results in only slightly diminished activity compared to THIQ itself. The relative activities of 4-8 were examined in terms of the conformational descriptors tau1 and tau2. Although there is no correlation between tau1 and activity as a PNMT inhibitor, a qualitative relationship between tau2 (endo or exo) and activity with PNMT is apparent. We believe that the binding of the N-H and/or N-lone pair of electrons may influence the spatial orientation of these molecules at the active site, resulting in positive binding interactions for compounds 4 and 8 and negative interactions for analogues 5-7. The results from the current investigation are compared to those obtained from a similar study involving conformationally defined amphetamines.

C-H functionalization of tertiary amines by cross dehydrogenative coupling reactions: Solvent-free synthesis of alpha-aminonitriles and beta-nitroamines under aerobic condition

A solvent-free synthesis of alpha-aminonitriles and beta-nitroamines by oxidative cross-dehydrogenative coupling under aerobic condition is reported.
