Two-dimensional covalent organic frameworks (2D-COFs) with permanent porosity, columnar pi-arrays and excellent stability, that can become an excellent platform for heterogeneous photocatalysis of organic transformations. Here we report a predesigned imine-based COF with electron donor and acceptor structure. And new framework possesses large surface area, high crystallinity, outstanding stability and broad absorption range in the visible-light region as well as good photoelectric response characteristics. Importantly, it was found to be a highly effective heterogeneous photocatalyst for reductive dehalogenation of phenacyl bromide derivatives and alpha-alkylation of aldehydes under irradiation of visible-light. In addition, the COF gave good recyclability and could be reused after a simple separation manipulation. The current present still reveals a great prospect for 2D-COFs as metal-free, heterogeneous photocatalysts for organic transformations.

The theoretical calculations showed that hyper-conjugation, the main underlying factor hindering the aerobic oxidation of tetrahydroisoquinolines, is relieved both by pi- and sigma-donating substituents. This mechanistic insight provided a novel photocatalytic route based on N-substituted auxiliaries that facilitated the conversion of tetrahydroisoquinolines into the corresponding isoquinolines in just three simple steps (yield 71.7% in bulk-solution phase), using unmodified Ru(bpy)3Cl2 photocatalyst, sun energy, atmospheric O2, and at ambient temperature.

Herein we describe the development and application of a “toolbox” of monoamine oxidase variants from Aspergillus niger (MAO-N) which display remarkable substrate scope and tolerance for sterically demanding motifs, including a new variant, which exhibits high activity and enantioselectivity toward substrates containing the aminodiphenylmethane (benzhydrylamine) template. By combining rational structure-guided engineering with high-throughput screening, it has been possible to expand the substrate scope of MAO-N to accommodate amine substrates containing bulky aryl substituents. These engineered MAO-N biocatalysts have been applied in deracemization reactions for the efficient asymmetric synthesis of the generic active pharmaceutical ingredients Solifenacin and Levocetirizine as well as the natural products (R)-coniine, (R)-eleagnine, and (R)-leptaflorine. We also report a novel MAO-N mediated asymmetric oxidative Pictet-Spengler approach to the synthesis of (R)-harmicine.
