Functionally selective G protein-coupled receptor ligands are valuable tools for deciphering the roles of downstream signaling pathways that potentially contribute to therapeutic effects versus side effects. Recently, we discovered both Gi/o-biased and beta-arrestin2-biased D2 receptor agonists based on the Food and Drug Administration (FDA)-approved drug aripiprazole. In this work, based on another FDA-approved drug, cariprazine, we conducted a structure-functional selectivity relationship study and discovered compound 38 (MS1768) as a potent partial agonist that selectively activates the Gi/o pathway over beta-arrestin2. Unlike the dual D2R/D3R partial agonist cariprazine, compound 38 showed selective agonist activity for D2R over D3R. In fact, compound 38 exhibited potent antagonism of dopamine-stimulated beta-arrestin2 recruitment. In our docking studies, compound 38 directly interacts with S1935.42 on TM5 but has no interactions with extracellular loop 2, which appears to be in contrast to the binding poses of D2R beta-arrestin2-biased ligands. In in vivo studies, compound 38 showed high D2R receptor occupancy in mice and effectively inhibited phencyclidine-induced hyperlocomotion.

This review, with over 600 references, summarizes the recent applications of photoredox catalysis for organic transformation and polymer synthesis. Photoredox catalysts are metallo- or organo-compounds capable of absorbing visible light, resulting in an excited state species. This excited state species can donate or accept an electron from other substrates to mediate redox reactions at ambient temperature with high atom efficiency. These catalysts have been successfully implemented for the discovery of novel organic reactions and synthesis of added-value chemicals with an excellent control of selectivity and stereo-regularity. More recently, such catalysts have been implemented by polymer chemists to post-modify polymers in high yields, as well as to effectively catalyze reversible deactivation radical polymerizations and living polymerizations. These catalysts create new approaches for advanced organic transformation and polymer synthesis. The objective of this review is to give an overview of this emerging field to organic and polymer chemists as well as materials scientists.

Hydroamination allows for the direct access to synthetically important amines. Controlling the selectivity of the reaction with efficient, widely applicable, and economic catalysts remains challenging, however. This paper reports an iron-catalyzed formal anti-Markovnikov hydroamination and hydroamidation of allylic alcohols, which yields gamma-amino and gamma-amido alcohols, respectively. Homoallylic alcohol is also feasible. The catalytic system, consisting of a pincer Fe-PNP complex (1-4 mol %), a weak base, and a nonpolar solvent, features exclusive anti-Markovnikov selectivity, broad substrate scope (>70 examples), and good functional group tolerance. The reaction could be performed at gram scale and applied to the synthesis of drug molecules and heterocyclic compounds. When chiral substrates are used, the stereochemistry and enantiomeric excess are retained. Further application of the chemistry is seen in the functionalization of amino acids, natural products, and existing drugs. Mechanistic studies suggest that the reaction proceeds via two cooperating catalytic cycles, with the iron complex catalyzing a dehydrogenation/hydrogenation process while the amine substrate acts as an organocatalyst for the Michael addition step.

Synthesis and preclinical evaluation of novel PET probes for P-glycoprotein function and expression

P-glycoprotein (P-gp) is an ATP-dependent efflux pump protecting the body against xenobiotics. A P-gp substrate (7) and an inhibitor (6) were labeled with 11C, resulting in potential tracers of P-gp function and expression. Methods: 6 and 7 were labeled using 11CH3I. 11C-verapamil was prepared as published previously, using 11C-methyl triflate. MicroPET scans (with arterial sampling) and biodistribution studies were performed in rats pretreated with saline, cyclosporin A (CsA, 50 mg/kg), or cold 6 (15 mg/kg). Results: The radiochemical yields of 11C-6 and 11C-7 were approximately 30%with a total synthesis time of 45 min. Cerebral distribution volumes (DV) of 11C-6 (2.35 ¡À 0.11) and 11C-7 (1.86 ¡À 0.15) in saline-treated rats were higher than of 11C-verapamil (0.64 ¡À 0.12). DVs of 11C-7 and 11C-verapamil were significantly increased by CsA (to 5.26¡À0.14 and 5.85¡À0.32, respectively). The DV of 11C-6 was reduced by cold 6 (to 1.65¡À0.03). Its uptakewas also reduced (up to 67%) in several peripheral organs that express P-gp. Conclusions: 11C-7 is a novel tracer of P-gp function with higher baseline uptake than 11C-verapamil. Upregulation of P-gp function in response to treatment (which is hard to detect with 11C-verapamil) may be detectable using 11C-7 and PET. Because 11C-6 shows specific binding in target organs, this compound is the first PET tracer allowing measurement of P-gp expression.
