A solution of 6-bromo-dihydroisoquinoline (300 mg, 1.42 mmol) in CH2Ch (8 mL) was cooled to 0 C and iPr2NEt (243 mg, 1.88 mmol) and CbzCl (322 mg, 1.88 mmol) were added. The reaction was stirred at room temperature forl6 h. The reaction was diluted with CH2CI2 (10 mL) and poured into water (20 mL). The layers were separated and the aqueous layer was extracted with CH2CI2 (3 x 20 mL). The combined organic layers were dried (Na2S04) and concentrated under reduced pressure. The crude mixture was purified via flash column chromatography eluting with EtOAc:hexanes (5:95) to give 400 mg (81%) of compound KTL- 01-202 as a clear oil: NMR (400 MHz) delta 7.41 – 7.27 (comp, 7 H), 6.97 (d, / = 10.3 Hz, 1 H), 5.18 (s, 2 H), 4.59 (s, 2 H), 3.70 (br s, 2 H), 2.82 (br s, 2 H); HRMS (ESI) m/z CivHieBrNCh (M+Na)+ calcd for 368.0257 and 370.0238; found 368.0257 and 370.0238., 226942-29-6

To a solution of 6- bromo-l,2,3,4-tetrahydroisoquinoline (4.19 g, 19.7 mmol) in DCM (75 mL) was added 4- fluoro-2-methylbenzaldehyde (3.0 g, 22 mmol) and acetic acid (1.13 mL, 19.7 mmol). Then sodium triacetoxyborohydride (5.4 g, 26 mmol) was added. The mixture was stirred at RT for 16 hrs. The mixture was quenched with water and extracted with DCM. The organic layer was washed with brine, dried over Na2S04 and concentrated. The residue was purified by recrystallization with EtOAc to give 6-bromo-2-(4-fluoro-2- methylbenzyl)-l,2,3,4-tetrahydroisoquinoline (3.4 g, 10.17 mmol, 51.5 % yield). LCMS (M+H) = 333.95 and 335.90.

To a dry microwave vial under nitrogen was added 2,4-dichloro-6-methyl-l,3,5-triazine (150 mg, 0.915 mmol), 6-bromo-l,2,3,4-tetrahydroisoquinoline (195 mg, 0.919 mmol) and anhydrous NMP (2.5 mL). The reaction was flushed with nitrogen, then treated with triethylamine (400 mu, 2.87 mmol), capped and allowed to stir at room temp for 30 min. The reaction was then treated with morpholine (800 mg, 9.18 mmol) and stirred at room temp for 2 h. The resulting white solid was collected by vacuum filtration to afford 4-(4- (6-bromo-3,4-dihydroisoquinolin-2(lH)-yl)-6-methyl-l,3,5-triazin-2-yl)mophiholine, 200 mg (54%). LCMS (M+l) = 390.1, 392.1. General procedure: The mixtures of 4-(bromomethyl)benzoic acid (200 mg, 0.93 mmol, 1 equiv.) and correspondingtetrahydroisoquinolines A (1.86 mmol, 2 equiv.) in 8 mL anhydrous THF were refluxed for 8 h,and the progress of the reaction was followed using TLC. After completion of the reaction, the reactionmixture was cooled to room temperature and the solvent was removed under vacuo. The resultantsolid was dissolved in 1N NaOH (20 mL) and then extracted with CH2Cl2 (3 20 mL). The aqueouslayer was acidified to pH = 1 using 10% HCl (20 mL), resulting in precipitation of the final compounds.

Step A: Preparation of l-(6-Bromo-3,4-dihydroisoquinolin-2(lH)-yl)-2- hydroxyethanone. A magnetically stirred flask equipped with a drying tube was charged with a solution of6-bromo-l,2,3,4-tetrahydroisoquinoline (3.43 g, 16.2 mmol) in anhydrous toluene (40 mL), to which was added 2,2-dimethyl-l,3-dioxolan-4-one (1.9 g, 16.2 mmol). The resulting solution was refluxed for 20 h. The reaction mixture was cooled, extracted with 1 N HCl (30 mL), followed by brine (20 mL), and the organic extract was dried over MgSO4. The resulting solution was reduced in volume to about 25 mL, and heptane (25 mL) was gradually added over 20 min as precipitate formed. The resulting white solid was collected by filtration and rinsed with 1 : 1 toluene/heptane to provide the title compound. LCMS m/z = 270.1 [M+H]+; 1H NMR (400 MHz, CDCl3) delta 2.48 (bs, IH), 2.87-2.94 (m, 2H), 3.55 (t, J= 5.9 Hz, 1.2H), 3.89 (t, J= 6.1 Hz, 0.8H), 4.26 (s, 2H), 4.40 (s, 0.8H), 4.75 (s, 1.2H), 7.00 (d, J= 8.3 Hz, 0.4H), 7.07 (d, J= 8.3 Hz, 0.6H), 7.32-7.39 (m, 2H).

General procedure: To the solution of amine (2a-10a) (1mmol) in DCM (5mL), triethylamine (2mmol) was added followed by benzylsulfonyl chloride, 2-Phenylethanesulfonyl chloride, benzene sulfonyl chloride, benzoyl chloride or phenylacetyl chloride (1.5mmol), and the mixture was stirred overnight at room temperature. The reaction was filtered, concentrated and solved by EA (20mL), then washed with ethyl acetate (3¡Á15mL). The organic was combined and was dried over Na2SO4.
