Quenching of 2 with Grignard reagents in tetrahydrofuran afforded the corresponding 1-alkyl and 1-aryl substituted tetrahydroisoquinolines (31 to 78%). Interestingly, nitrile 10 reacted very rapidly (<2 min at 0C) with phenylmagnesium bromide to give 2-methyl-1-phenyl-1,2,3,4-tetrahydroisoquinoline (3, 100%), but failed to react with excess phenyllithium even at 20C. A series of zatebradine analogues, differing in the basic moiety and in the methylene spacer, have been synthesized; their negative chronotropic activity has been determined in guinea pig atria. The most active compounds have been studied for their blocking properties on the hyperpolarization-activated current If (which is one of the main currents underlying automatic activity in the sinus node) measured on ventricular myocytes of old spontaneously- hypertensive rats (SHR) by means of the patch-clamp technique. The majority of the substances were able to block If, with one of them (15) being slightly more potent than zatebradine. Diseases of the CNS are often complex and involve multiple receptor systems and thus, the treatment options for these diseases must focus on targeting the multiple receptors implicated in the various disorders. In our previous campaign to find multi-receptor ligands, we have identified the benzothiazole 1a as an initial lead molecule. In the current work, we have expanded the structure affinity relationship (SAFIR) of 1a resulting in the identification of a partially restrained butyrophenone 3j as a potent and selective dual 5-HT1Aand 5-HT7receptor ligand. It is expected that compound 3j may serve as a new lead for further development in our search for newer and novel ligands with the potential to treat diseases of CNS origin.
