A metal-free heterogeneous photocatalytic system has been developed for highly efficient benzylic C-H oxygenations using oxygen as an oxidant. This visible light mediated oxidation reaction utilizes graphitic carbon nitride (g-C3N4) as a recyclable, nontoxic and low cost photocatalyst. Mild reaction conditions allow for the generation of synthetically and biologically valued isochromannones, phthalides, isoquinolinones, isoindolinones and xanthones from readily accessible alkyl aromatic precursors in good yields. The heterogeneous nature of the g-C3N4 catalytic system enables easy recovery and recycling as well as the use in multiple runs without loss of activity. The synthetic utility of this “green” methodology was further demonstrated by applying in bioactive and drug valued target syntheses.

Similarly, potassium cyanide in acetonitrile provided 1-cyano-2-methyl-1,2,3,4-tetrahydroisoquinoline (10, 77%). Quenching of 2 with Grignard reagents in tetrahydrofuran afforded the corresponding 1-alkyl and 1-aryl substituted tetrahydroisoquinolines (31 to 78%). Interestingly, nitrile 10 reacted very rapidly (<2 min at 0C) with phenylmagnesium bromide to give 2-methyl-1-phenyl-1,2,3,4-tetrahydroisoquinoline (3, 100%), but failed to react with excess phenyllithium even at 20C. In the current work, we have expanded the structure affinity relationship (SAFIR) of 1a resulting in the identification of a partially restrained butyrophenone 3j as a potent and selective dual 5-HT1Aand 5-HT7receptor ligand. It is expected that compound 3j may serve as a new lead for further development in our search for newer and novel ligands with the potential to treat diseases of CNS origin.

Only 5 isoquinoline derivatives produced 50% inhibition of [3H]dopamine uptake (IC50 = 8.0-50.0 microM), none of which were as potent as MPP+ (IC50 = 0.33 microM). These findings suggest that isoquinoline derivatives are moderate to poor substrates for the dopamine reuptake system and that high concentrations of, or prolonged exposure to, isoquinoline derivatives may be necessary to cause neurodegeneration.
