We have designed and synthesized a series of indole-based sigma2 receptor ligands containing 5,6-dimethoxyisoindoline or 6,7-dimethoxy-1,2,3,4-tetrahydroisoquinoline as pharmacophore. In vitro competition binding assays showed that all ten ligands possessed low nanomolar affinity (Ki?=?1.79?5.23?nM) for sigma2 receptors and high subtype selectivity (Ki (sigma2)/Ki (sigma1)?=?56?708). Moreover, they showed high selectivity for sigma2 receptor over the vesicular acetylcholine transporter (>1000-fold). The corresponding radiotracers [18F]16 and [18F]21 were prepared by an efficient one-pot, two-step reaction sequence with a home-made automated synthesis module, with 10?15% radiochemical yield and radiochemical purity of >99%. Both radiotracers showed high brain uptake and sigma2 receptor binding specificity in mice.

Synthesis and preclinical evaluation of novel PET probes for P-glycoprotein function and expression

P-glycoprotein (P-gp) is an ATP-dependent efflux pump protecting the body against xenobiotics. A P-gp substrate (7) and an inhibitor (6) were labeled with 11C, resulting in potential tracers of P-gp function and expression. Methods: 6 and 7 were labeled using 11CH3I. 11C-verapamil was prepared as published previously, using 11C-methyl triflate. MicroPET scans (with arterial sampling) and biodistribution studies were performed in rats pretreated with saline, cyclosporin A (CsA, 50 mg/kg), or cold 6 (15 mg/kg). Results: The radiochemical yields of 11C-6 and 11C-7 were approximately 30%with a total synthesis time of 45 min. Cerebral distribution volumes (DV) of 11C-6 (2.35 ¡À 0.11) and 11C-7 (1.86 ¡À 0.15) in saline-treated rats were higher than of 11C-verapamil (0.64 ¡À 0.12). DVs of 11C-7 and 11C-verapamil were significantly increased by CsA (to 5.26¡À0.14 and 5.85¡À0.32, respectively). The DV of 11C-6 was reduced by cold 6 (to 1.65¡À0.03). Its uptakewas also reduced (up to 67%) in several peripheral organs that express P-gp. Conclusions: 11C-7 is a novel tracer of P-gp function with higher baseline uptake than 11C-verapamil. Upregulation of P-gp function in response to treatment (which is hard to detect with 11C-verapamil) may be detectable using 11C-7 and PET. Because 11C-6 shows specific binding in target organs, this compound is the first PET tracer allowing measurement of P-gp expression.

As a result, the stronger hydrogen bonds formed by the neutral molecules play a secondary role in the crystal structure formation.

Alkaloid constituents from flower buds and leaves of sacred lotus (Nelumbo nucifera, Nymphaeaceae) with melanogenesis inhibitory activity in B16 melanoma cells

Methanolic extracts from the flower buds and leaves of sacred lotus (Nelumbo nucifera, Nymphaeaceae) were found to show inhibitory effects on melanogenesis in theophylline-stimulated murine B16 melanoma 4A5 cells. From the methanolic extracts, a new alkaloid, N-methylasimilobine N-oxide, was isolated together with eleven benzylisoquinoline alkaloids. The absolute stereostructure of the new alkaloid was determined from chemical and physicochemical evidence. Among the constituents isolated, nuciferine, N-methylasimilobine, (-)-lirinidine, and 2-hydroxy-1-methoxy-6a,7-dehydroaporphine showed potent inhibition of melanogenesis. Comparison of the inhibitory activities of synthetic related alkaloids facilitated characterization of the structure-activity relationships of aporphine- and benzylisoquinoline-type alkaloids. In addition, 3-30 muM nuciferine and N-methylasimilobine inhibited the expression of tyrosinase mRNA, 3-30 muM N-methylasimilobine inhibited the expression of TRP-1 mRNA, and 10-30 muM nuciferine inhibited the expression of TRP-2 mRNA.
