An efficient and operationally simple synthesis of tetrahydrofuran-derived spiro-beta-lactams using the ketene-imine cycloaddition route is described. Also the preparation of spiro-N-sulfonyl-beta-lactam derivatives, which are analogs of monobactams, is reported. As far as we know, this is the first time that an unsymmetrical cyclic ketene is used in a Staudinger-type reaction. The experimental evidence suggests the involvement of a ketene derived from the acyl chloride precursor in the reaction. High-level ab initio calculations have been performed in order to get insight into the electronic effects controlling the stereochemical outcome of the reactions.

Interestingly, these compounds were more potent against breast carcinoma cell lines (MCF-7 and T-47D) than the lymphoid origin tumor cell lines (Jurkat and BHL-89). In particular, the (S)-proline derivative 10 exhibited preferential cytotoxicity to adherent cells (IC50 = 0.5 muM), indicative of better potential in blocking the growth of solid tumors rather than the disseminated ones.

The esters include the propionate, butyrate, methoxyacctate, beta-methoxypropionate, 4-methoxybutyrate, 2-tetrahydrofuroate, 2-furoate, (N,N-dimethylamino)acetate, (methylthio)acetate, 2- and 4-pyridine-carboxylates, 2-pyridylacetate, 4-pyridylacetate, phenylacetate, andp-methoxy-,p-chloro-, and p-(trifluoromethyl)phenylacetates. The rates and kinetic orders of the reactions of 3,5-dimethylphenyl propionate in various solvents at 35C gave the following second-order rate constants (104k2, L mol-1 sec-1) for the following major aggregate species: THF tetramer, 6.5; DME tetramer, 3.3 (40C); dioxolane, 13, hexamer, 71; pyridine tetramer, 2.2, dimer, 29. For 3,5-dimethylphenyl beta-methoxypropionate, the order of reactivity is dioxolane > DME > THF. These results are interpreted in terms of a preequilibrium in which a solvent on lithium in the tetramer is replaced by the ester. The rates of transesterification have been compared with the rates of hydrolysis in 30% aqueous ethanol for the above series of esters. Those esters that have a second Lewis base center proximal to the ester function show significantly increased reactivity in transesterification, which is attributed to a complex-induced proximity effect.

Binding assays with rat muscarinic receptor subtypes revealed that the quinuclidin-3-yl 1-aryl-1,2,3,4- tetrahydroisoquinoline-2-carboxylate derivatives showed high affinities for the M3 receptor, and selectivity for the M3 receptor over the M2 receptor. Of these derivatives, (+)-(1S,3’R)-quinuclidin-3′-yl 1-phenyl-l,2,3,4-tetrahydroisoquinoline-2-carboxylate monohydrochloride (9b) exhibited almost the same inhibitory activity against bladder contraction to that of oxybutynin (1), and more than 10-fold selectivity for bladder contraction versus salivary secretion, demonstrating that 9b may be useful for the treatment of symptoms associated with overactive bladder without having side effects such as dry mouth.
