General procedure: To a solution of bromoacetyl chloride (1.40g, 8.17mmol) in10mL of dry THF cooled at 0C, under argon atmosphere, was added dropwise, under vigorous stirring, a solution of the appropriate amine (5.45mmol) and triethylamine (0.276g, 2.72mmol) in dry THF (10mL). After 30min at 0 and 30min at room temperature the reaction mixture was quenched by the addition of 50ml of H2O and extracted with dichloromethane (3¡Á50ml). The combined organic extracts were washed with saturated NaHCO3 and brine, and then dried over anhydrous Na2SO4. Evaporation under reduced pressure gave the crude 3-bromopropanamide derivatives (1-3) which were purified by flash chromatography on silica gel, using CH2Cl2 and cyclohexane as eluent.

The crude product was purified by preparative HPLC with a C 18 column using acetonitrile: water (with 0.1% formic acid) as eluant with a gradient from 10:90 (v/v) to 80:20 (v/v) at a flow rate of 45 mL/min to give methyl 2-(6,7,8-trimethoxyquinazoline-4-yl)- l,2,3,4-tetrahycroisoquinoline-8-carboxylate.

The resulting solution was heated and stirred overnight under reflux. The resulting solution was extracted with ethyl acetate, washed with water and saturated aqueous solution of sodium chloride, and then dried with anhydrous sodium sulfate. After separating the desiccant by filtration, the product was purified by silica gel column chromatography to obtain 1.1 g of 2-(2-(ethoxycarbonyl)ethyl)-7-nitro-1,2,3,4-tetrahydroisoquinoline (yield: 57percent, oily product).

The residue was purified by flash column chromatography eluting with DCM / EtOAc: 8 / 2 to give the title compound as a yellow oil (25 mg, 15%). LC/MS: m/z 498 (M+H)+, Rt : 3.83 min.

The solvent was removed under a gentle stream of niotrogen and the crude material was purified via silica gel chromatography (40g SiCh column, dichloromethane: ethyl acetate 100:0 -> 0: 100) to afford 2-(l-(2-(6-bromo-3,4-dihydroisoquinolin-2(lH)-yl)pyrimidin-4-yl)piperidin-4- yl)propan-2-ol, 2.0 methyl-2-pyrrolidinone, 642.2 mg (48%). LCMS (M+l) = 431.2 and 433.1.

The solution was stirred 10 minutes with 1 N NaOH, and the layers were separated. Paraformaldehyde (2.217 g, 73.8 mmol), acetic acid (4.23 ml, 73.8 mmol) and sodium cyanoborohydride (4.64 g, 73.8 mmol) were added. The reaction was heated at 90 0C overnight. The reaction was cooled to room temperature, and quenched with saturated aqueous sodium bicarbonate. The layers were separated, and the organic layer was dried over MgSO4, filtered, and concentrated onto silica gel. The reaction was purified by flash chromatography (50percent ethyl acetate:hexanes for 20 minutes, then to 100percent ethyl acetate:hexanes over 30 minutes) to provide the title compound. MS (DCI) m/e 193 (M+H)+.
